Immunotherapy to deplete key Treg subsets in solid tumors

Summary
Aim 1: Compare the effect of SON-211 and SON-301 monotherapies vs. no therapy on the 10 subsets of cancer-infiltrating regulatory T cells. scRNAseq will be performed on purified lymphocytes from flow- sort purified B16 tumors to evaluate for the 10 Treg subsets.6 Specifically, we will determine if SON-211 is effective in eliminating the highly suppressive OX40hi GITRhi Treg subset.
Aim 2: Compare the effect of SON-211/Anti-PD1 and SON-301/Anti-PD1 dual therapies vs. each monotherapy vs. no therapy. scRNAseq will be performed on purifed lymphocytes from flow-sort purified B16 tumors subjected to treatment to determine if dual therapy with Anti-PD1 is effective in eliminating the highly suppressive OX40hi GITRhi Treg subset and expanding the Th1-like TBX21 [Tbet], IFNg Treg subset.
