Cells are harvested from a suspension culture, washed and resuspended in the developmental buffer (DB: 2 mM MgCl2, 0.2 mM CaCl2 in 10 mM Na/K phosphate pH 6.5) at 5x106 cells /ml. 5 ml cells are then spotted on DB agar containing cAMPγS at concentrations of 0, 10-6, 10-5 M. cAMPγS should be mixed with the melted DB agar when the agar is at about 40 oC. Terminal phenotypes can be observed within 36 hours.
